Targeted protein degradation (TPD) is an emerging research strategy centered around using small molecules called “degraders” to trigger ubiquitin-dependent degradation of specific proteins. This approach shows promise for oncology drug development for many reasons, including the ability to selectively disrupt critical signaling pathways, target previously inaccessible cancer-causing proteins, and overcome drug resistance. Degradation-based strategies also complement basic genetic experiments while offering better target selectivity and enabling functional studies with superior kinetic resolution. The unique properties of single-variable domain or heavy-chain only antibodies have revolutionized how researchers exploit antibodies to resolve essential questions in life science research and diagnostics, and even generate novel therapeutics. Seen in animals such as camels and sharks, heavy chain-only antibodies are modified to generate VHH antibodies which are small antibody fragments comprising only the variable heavy chain. Weighing in at only 12 kDA–15 kDA, these nanobodies are one-tenth the mass of conventional antibodies (150 kDA–160 kDA). SpeakerS: Natalie Castellana, Ph.
